Structure and function of reconstituted AMPA receptors

چکیده
Background: Ionotropic glutamate receptors mediate fast excitatory synaptic transmission in the vertebrate CNS. Results: Conformational flexibility and dimensions of functional membrane-embedded full-length GluA2flop receptors are characterized by atomic force microscopy. Conclusion: Conformational flexibility and dimensions are both strongly affected by receptor density. Significance: The reconstitution protocol lays the foundation for correlated structure-function analysis of membrane-embedded glutamate receptors.

Single channel recordings of reconstituted AMPA receptors reveal low and high conductance states.
Glutamate receptors belonging to the AMPA (alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid) subclass were partially purified 30- to 60-fold from forebrain of adult rats and incorporated into planar bimolecular lipid membranes. The channel conductance associated with the reconstituted receptors was activated by kainate and AMPA in a manner that suggests cooperative binding of two to thr...
متن کاملEffects of heparin on the properties of solubilized and reconstituted rat brain AMPA receptors.
Heparin was found to bind to alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA) receptors and to alter their functional properties. AMPA receptors solubilized in 0.4% Triton X-100 bound to a heparin-agarose column and were eluted by 0.4 M NaCl. Soluble heparin inhibited 10 nM [3H]AMPA binding to detergent-solubilized receptors by 75% (IC50 = 10 micrograms/ml), but had little effect...
متن کاملRole of the AMPA receptors of paragigantocellularis lateralis nucleus in the inflammatory pain modulation in male rat
Introduction: The 17β-estradiol acts as a neurosteroid in the brain and modulates nociception by binding to the estrogen receptors and also by allosteric interaction with other membrane-bound receptors like glutamate receptors. Paragigantocellularis lateralis nucleus (LPGi) is one of the important brain regions implicated in the pain modulation. So, this study was designed to evaluate the ...

Background: Environmental signals have an essential role in the maturation of neural circuits during critical period of brain development. It has been shown that, change in visual signals during critical period of brain development changes structure and function of glutamate receptors in the visual cortex. After processing in visual cortex, part of visual signals goes to the hippocampus and mak...

Background and purpose: The paragigantocellularis lateralis nucleus (LPGi) is one of the important nuclei involved in the noradrenergic descending pain modulation. 17β-Estradiol modulates nociception by binding to the estrogen receptors and also by allosteric interaction with other membrane-bound receptors like the glutamate receptors. In this study, the role of AMPA receptors of the LPGi ...
